Fragment E was ligated with plasmid PBR322 and then transformed into E. coli JM109 host cells. Plasmid DNA was isolated from subclones, and then DNA was sequenced according to Maxam and Gilbert procedure. The sequence of this fragment was obtained through labelling at the ECOR1 site and Hind III site. Thus, base sequencing gave us a chance to look at the same part of amino acid sequence of α-subunit of fatty acid synthatase. Since the size of the fragment E is approximately 850 base pairs it could code for 283 amino acids. This might show that there was no active site on fragment E coding for anyone of the known enzymes of the fatty acid synthetase system. When this amino acid sequence was compared to the amino acid sequence of the enzymes of fatty acid synthetase system, no homology was detected.
